RESUMO

DNA has emerged as an appealing material for information storage due to its great storage density and durability. Random reading and rewriting are essential tasks for practical large-scale data storage. However, they are currently difficult to implement simultaneously in a single DNA-based storage system, strongly limiting their practicability. Here, a "Cell Disk" storage system is presented, achieving high-density in vivo DNA data storage that enables both random reading and rewriting. In this system, each yeast cell is used as a chamber to store information, similar to a "disk block" but with the ability to self-replicate. Specifically, each genome of yeast cell has a customized CRISPR/Cas9-based "lock-and-key" module inserted, which allows selective retrieval, erasure, or rewriting of the targeted cell "block" from a pool of cells ("disk"). Additionally, a codec algorithm with lossless compression ability is developed to improve the information density of each cell "block". As a proof of concept, target-specific reading and rewriting of the compressed data from a mimic cell "disk" comprising up to 105 "blocks" are demonstrated and achieve high specificity and reliability. The "Cell Disk" system described here concurrently supports random reading and rewriting, and it should have great scalability for practical data storage use.

In today's society, people's lives are increasingly inseparable from computer information. Due to the continuous improvement of technology and the rapid development of internet technology, the network environment is becoming more and more complex, which makes it easy to cause loopholes in the information retrieval system when people use the network. Therefore, it is especially important to search for legal knowledge by computer. In order to adapt to this change and demand, we need a retrieval system to provide the corresponding search function, legal information content, and management and other services, so as to achieve the purpose of computer legal information retrieval. The legal information retrieval system is computer based, draws conclusions from the analysis of relevant data, and then applies them to judicial trial cases, criminal investigations, and other fields to provide a reference for relevant legal issues. The system is designed to combine computer technology with a criminal investigation and other fields, and then analyze the data to draw the corresponding conclusions. The retrieval algorithms used are mainly image and content retrieval algorithms, and image retrieval algorithms mainly use image segmentation technology, while content retrieval algorithms mainly use the cuckoo algorithm. At present, the information construction and economic and social development in China have become one of the issues of common concern and need to be solved by all countries in the world. The study of the legal information retrieval system is of great importance in the construction of information technology and the development of economic society.

Exploring effective electrocatalysts for oxygen evolution reaction (OER) is a crucial requirement of many energy storage and transformation systems, involving fuel cells, water electrolysis, and metal-air batteries. Transition-metal oxides (TMOs) have attracted much attention to OER catalysts because of their earth abundance, tunable electronic properties, and so forth. Defect engineering is a general and the most important strategy to tune the electronic structure and control size, and thus improve their intrinsic activities. Herein, OER performance on spinel CuCo2O4 was greatly enhanced through cation substitution and size reduction. Ce-substituted spinel CuCeδCo2-δOx (δ = 0.45, 0.5 and 0.55) nanoparticles in the quantum dot scale (2-8 nm) were synthesized using a simple and facile phase-transfer coprecipitation strategy. The as-prepared samples were highly dispersed and have displayed a low overpotential of 294 mV at 10 mA·cm-2 and a Tafel slope of 57.5 mV·dec-1, which outperform commercial RuO2 and the most high-performance analogous catalysts reported. The experimental and calculated results all confirm that Ce substitution with an appropriate content can produce rich oxygen vacancies, tune intermediate absorption, consequently lower the energy barrier of the determining step, and greatly enhance the OER activity of the catalysts. This work not only provides advanced OER catalysts but also opens a general avenue to understand the structure-activity relationship of pristine TMO catalysts deeply in the quantum dot scale and the rational design of more efficient OER catalysts.

AIMS: Expression of gonadotropin-releasing hormone receptor (GnRHR) has been demonstrated in a number of malignancies. The aim is to investigate the expression of GnRHR and prognosis in gastric cancer. METHODS AND MATERIALS: GnRHR mRNA was examined in tumor and non-tumor tissues from 48 gastric cancer patients by Real-time PCR. The GnRHR protein expression was performed by immunohistochemical analysis. RESULTS: The expression of GnRHR mRNA was higher (mean ± SD, -10.06 ± 1.28) in gastric tumor tissues than matched non-tumor tissues (mean ± SD, -12.43 ± 1.33). GnRHR mRNA expression was associated with lymph node metastasis, distant metastasis, and TNM stage. We found the decreased expression of GnRHR mRNA were significantly correlated with poor overall survival (P = 0.003). Immunocytochemical staining of GnRHR in tumor tissues showed mainly weak staining (43.48%, 10/23) and moderate staining (21.74%, 5/23) in high GnRHR mRNA patients, and mainly negative staining in low GnRHR mRNA patients. And the staining of GnRHR was not detection in tumor tissues for more than half of gastric patients (52.08%, 25/48). These results implied that the loss of GnRHR protein could be a main event in gastric cancer. CONCLUSION: The GnRHR expression is very low in gastric cancer, and the loss of GnRHR expression could be a poor prognostic factor, which implied that GnRHR could play an important role in the development of gastric cancer.

N-[(2-Hydroxy-3-trimethylammonium)propyl] chitosan chloride (HTCC) was chemically modified using glycidyltrimethylammonium chloride (GTMAC). A new composite hydrogel was prepared using the mixture of HTCC and α-ß-glycerophosphate (α-ß-GP). The gelation of HTCC/GP mainly depended on the concentration and proportion of HTCC and GP. Thermogravimetric analysis exhibited high stability of HTCC/GP hydrogels. Surface morphology assay demonstrated that HTCC/GP hydrogels were well constructed with three-dimensional (3D) porous structures in the range of 5 of 40 µm. The insulin was entrapped during the formation of hydrogel. In vitro, the insulin release was controlled by modifying the composition, drug loading, and pH condition. The hydrogel dissolved and released drug quickly under acidic condition, whereas it absorbed water and released drug slowly under neutral or basic conditions. The hydrogels were biocompatible, and the cells could adhere to and then migrated to the hydrogels. Furthermore, these cells were viable and retained 3D morphology inside the hydrogels. Interestingly, HTCC/GP hydrogel showed both thermo- and pH-sensitive properties. There are potential applications in tissue engineering, cell encapsulation, and intelligent drug delivery systems.

OBJECTIVE: To explore the associations between the single nucleotide polymorphism of human mismatch repair gene hMLH1 and the papillary thyroid carcinoma (PTC) in Chinese Han people. METHODS: A hospital based 1:1 matched case-control study was carried out. The single nucleotide polymorphism (-93G > A, 1151T > A and 655A > G) for 204 pairs of cases with PTC as well as healthy controls was identified by PCR-RFLP, PCR-ASO and DNA sequencing. RESULTS: With univariate analysis, we found that compared to 1151TT genotype, the TA genotype could increase the PTC risk marginally, with odds ratio (OR) of 2.15 (95%CI: 0.99 - 4.85); While the mutant genotype TA + AA could increase the PTC risk statistically significant, with OR of 2.15(95%CI: 1.02 - 4.69). With 2 x 4 cross-over study, we found that compared to -93GG and 1151TT genotypes, individuals with both -93GA + AA and 1151TA + AA could increase the PTC risk marginally, with OR of 2.50 (95%CI: 0.96 - 6.67); While, compared to 655AA and 1151TT genotypes, individuals with both 655AA and 1151TA + AA could increase the PTC risk statistically significant, with OR of 2.50 (95%CI: 1.02 - 4.73). Multivariate and conditional logistic regression analysis showed the genotype of 1151TA, the history of receiving CT diagnosis, the history of tumor, the negative life events and eating seafood frequently could increase the risk of PTC, with OR of 6.79 (95%CI: 3.18 - 14.49), 3.35 (95%CI: 1.93 - 5.80), 39.03 (95%CI: 3.70 - 41.60) and 3.98 (95%CI: 1.81 - 8.73); While, eating fruit frequently could decrease the PTC risk. CONCLUSION: The 1151TA + AA genotype, the history of receiving CT diagnosis, the history of tumor, the negative life events and eating seafood frequently were the risk factors of PTC, while eating fruit frequently was the protective factor.

OBJECTIVE: To understand the risk factors of primary angle-closure glaucoma. METHODS: One to one matched case-control study was conducted in this study. One hundred and ninety two PACG cases and 192 controls, matched by age and gender, were collected from Department of Ophthalmology, Affiliated Hospital of Nantong University. All of the participants were investigated for their demographic information, behavioral habits, disease history, glaucoma family history and received clinical ocular examinations. The difference between these two groups was analyzed. RESULTS: Several factors, including hypertension (OR = 2.004, P = 0.009), glaucoma family history (OR = 6.726, P = 0.003), presbyopia (OR = 3.192, P = 0.031), shallow anterior chamber (OR = 12.804, P = 0.000) and high cup-to-disc ratio (OR = 9.401, P = 0.007) were associated with PACG by multiply Logistic regression. The results did not support that smoking, drinking, myopia, diabetes mellitus and blood style were related to PACG. CONCLUSION: Follow up the populations with glaucoma family history, shallow anterior chamber or high cup-to-disc ratio are the main procedures for the decrease of incidence of PACG.

OBJECTIVE: To explore the relationship of the genetic polymorphisms and the haplotypes in hMLH1 and hMSH3 gene with the risk of papillary thyroid carcinoma (PTC) in Chinese Hans. METHODS: A hospital based 1:1 matched case-control study was carried out. The polymorphisms for 204 pairs of PTC cases and healthy controls were identified by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and allele specific oligonucleotide (PCR-ASO) assays. RESULTS: (1) The PTC risk was marginally increased in the hMLH1 1151TA genotype, with odds ratio (OR) of 2.15 (95%CI: 0.99-4.85); the PTC risk was significantly increased in the mutant genotype 1151TA+AA, with OR of 2.15 (95%CI: 1.02-4.69); (2) The haplotypes of -93G, 1151A, 655A in the hMLH1 gene could increase the PTC risk, with OR of 2.67 (95%CI: 1.16-6.53, P=0.011), compared with the haplotype of -93G, 1151T, 655A; (3) Compared to 3124A, 2835G haplotype in hMSH3 gene, the 3124G, 2835A haplotype could increase the PTC risk marginally, with OR of 3.08 (95%CI: 0.92-13.25). CONCLUSION: The 1151T/A polymorphism in hMLH1 was associated with PTC; both the haplotype of -93G, 1151A, 655A in hMLH1 and the 3124G, 2835A haplotype in hMSH3 were associated with PTC.

OBJECTIVE: To introduce the preparation and quality control of Solanum lyratum Extract Thunb gel. METHODS: The gel was prepared with Solanum lyratum Thunb Extract as main component and carbopol 940 as base material, and Solasodine was determined by TLCS. RESULTS: The preparation of this gel was feasible, its quality was reliable, its detection method was rapid, stable, and could be used to control the quality of Solanum lyratum Thunb Extract gel. CONCLUSIONS: Studies on Solanum lyratum Thunb Extract gel will find a new path for the treatment of eczema with mono traditional Chinese medicine.

OBJECTIVE: The quality control method of "Lü Kang Yin Xing Ye Pian" (LKYXYP) was studied and established. METHOD: The ginkgolides in LKYXYP were identified by TLC. The compounds of fatty acids in Perillae oil were determined by CC/MS. Flavones, ginkgolides and ginkgolic acids were determined by HPLC. RESULT: There were good linears relationship between the peak area and concentration, r = 0.999, RSD = 1.2% - 2.5% (n = 5), and recoveries of each composition was above 95%, the contation was stable in 24h. CONCLUSION: This method is proved to be accurate and able to be applied for the quality control of this preparation.
